either by predictions made by FTDNA based on
reported Y-DNA test results or as specifically
confirmed by an SNP test. Observations
of thousands of test results now indicate that some
Markers change or mutate at a faster rate than
others. While the actual faster rate has not
as yet been definitively calculated, not all markers
should be treated the same for evaluation purposes.
The DYS Markers displayed in
RED have shown a faster mutation rate then
the average; therefore, these markers are sometimes
very helpful at splitting lineages into sub sets, or
branches within the family tree. |

| NOTES:
The specific
direct ancestor (father, grandfather) for
New York Pro #02b (Squire
Knowles, b 1788) has not been determined via conventional
genealogical research. DNA test results for two other Knowles
Progenitors; i.e., Rhode Island
PRO #01 (Henry Knowles, b 1609) and
Massachusetts PRO #01
(Richard Knowles, b 1614) show close Y-DNA Haplotypes when compared
with New York Pro #02b.
Only one descendant of Rhode
Island PRO #01 (Henry Knowles, b 1609) has been tested to date
(Kit #09601). Therefore, a Haplotype (or Y-profile) for the
Rhode Island PRO #01 Knowles
Progenitor has not been verified (two or more participants are
required to establish and verify a Haplotype).
The test
results posted so far could mean that these three Knowles
Progenitors have a common direct male ancestor, although additional
test results (more participants and more markers for the
participants already tested) will be required.
The specific
notes below describe the Y-DNA profile comparisons among the
participants for these three Progenitors.
1) Test results for
the Rhode Island PRO #01 (Kit #09601) participant produced a near perfect match (11 for 12) at
the 12-marker level
with participants for New York PRO #02B
(Kits #09240 & 71419). The 25-marker results for
these two progenitors are
a 22 for 25 match (genetic distance of 3) and the 37-marker results are only 32
for 37 (genetic distance of 5), so the participants for these two
progenitors who are near matches at the 12-marker level may or
may NOT
share a common Knowles ancestor (additional testing to
67-markers may be warranted for Kit # 09601 to establish a
definitive connection between these two Knowles lines).
2) Test
results for a Massachusetts PRO
#01 descendant (Kit #97299) is also shown to be a close match
(11 for 12) with the Rhode Island PRO #01
descendant and an exact match (25 for 25) with the New York PRO #02b descendants. This
indicates that New York PRO #02b
is most likely a descendant of Massachusetts PRO
#01. These results
could also mean that all three of these Knowles lines share a common
direct male ancestor. Unfortunately, testing of more markers
will be required for Kits #09601 and #97299 to better establish the
relationship among these lines. Ultimately, all of these men
may have to be tested for 67 markers.
3) Test
results for a Massachusetts PRO
#01 descendant (Kit #93679) is also shown to be a close match
(11 for 12) and (23 for 25) with the New York PRO #02b
descendants (Kits #09240 & 71419). This a further indication
that the New York PRO #02b
is a descendant of Massachusetts PRO
#01. Additional testing (more markers) will help define
where the connection occurs.
4)
The test results at 37 markers for the participant representing New
York PRO #18 (Kit #127470) have eliminated the possibility there is
a direct male connection with the MA-01, NY-01a and NY-02b Knowles
lines. These results also indicate that NY-18 and RI-01 do
share a common male ancestor (GD=1 at 37 markers).
